Scenes will be filmed in the Utah desert for a story set in the late '60s in which the Doctor, Amy and Rory find themselves on a secret summons to the Oval Office.
The episodes have been written by new series boss Steven Moffat and co-produced with BBC America.
Production starts in Cardiff this month with Matt Smith and Karen Gillan.
Alex Kingston will reprises her role as River Song.
Moffat said: "The Doctor has visited every weird and wonderful planet you can imagine, so he was bound get round to America eventually.
"And of course every Doctor Who fan will be jumping up and down and saying he's been in America before. But not for real, not on location - and not with a story like this one."
It has been announced previously that series six has been split into two blocks, with the first airing on BBC One in spring 2011 and the second block showing in autumn 2011.

I'm just happy to hear we'll have another 2-parter with River Song - I've grown to really enjoy that character and I think SM should drag out the reveal of who she is as long as possible - she's very good - very interesting.
Mr. Moffat: Doctor Who has indeed been in America before. For real, and on location.
I realize some Whovians dislike the Fox film (which was shot in San Francisco and Vancouver). I've even heard people try to claim that the Eighth Doctor was not canon.
But watch "The Next Doctor." Watch "The Eleventh Hour." See that guy whose face is shown between Sylvester McCoy and Christopher Eccleston in the recaps?
The Fox film and Paul McGann's Eighth Doctor are canon. Deal with it. The Doctor has been in America.
calm down Dear, I think what Mr MOffatt meant was that the TV show has never filmed in the States. Paul McGann is no question a canonical Doctor, even prior to the clip in the Next Doctor I'd have said so as he regenerated from the 7th. But, as I said, the TV show didn't film there, FOX did a movie. Had a series been picked up that's a different story and one which, unfortunatley for Paul McGann was never picked up.
I would like to see Paul McGann in the new show, perhaps in a 50th Anniversary Multi Doctor special.
